The combination of alteplase, a recombinant tissue plasminogen activator, and edaravone, an antioxidant, reportedly enhances recanalization after acute ischemic stroke. We examined the influence of edaravone on the thrombolytic efficacy of alteplase by measuring thrombolysis using a newly developed microchip-based flow-chamber assay. Rat models of embolic cerebral ischemia were treated with either alteplase or alteplase-edaravone combination therapy. The combination therapy significantly reduced the infarct volume and improved neurological deficits. Human blood samples from healthy volunteers were exposed to edaravone, alteplase, or a combination of alteplase and edaravone or hydrogen peroxide. Whole blood was perfused over a collagen- and tbromboplastin-coated microchip; capillary occlusion was monitored with a video microscope and flow-pressure sensor. The area under the curve (extent of thrombogenesis or thrombolysis) at 30 minutes was 69.9% lower in the edaravone-alteplase- than alteplase-treated group. The thrombolytic effect of alteplase was significantly attenuated in the presence of hydrogen peroxide, suggesting that oxidative stress might hinder thrombolysis. D-dimers were measured to evaluate these effects in human platelet-poor plasma samples. Although hydrogen peroxide significantly decreased the elevation of D-dimers by alteplase, edaravone significantly inhibited the decrease. Edaravone enhances alteplase-mediated thrombolysis, likely by preventing oxidative stress, which inhibits fibrinolysis by alteplase in thrombi.

マイクロミニブタに8週間腸溶性ラクトフェリン(eLF)を経口摂取させたところ、高脂肪高コレステロール食(HFCD)による血清の総コレステロール値とLDL-コレステロール値の上昇が著しく減弱したが、HDL-コレステロールは変化しなかった。血管の組織検査より、eLFにより全身の粥状動脈硬化の進行が阻害されていた。eLF投与に特徴的な肝臓の遺伝子による経路分析では、ステロイド生合成経路に有意な変化が起こり、これらの遺伝子はすべて上方制御されていた。すなわち、HFCDにより阻害されたコレステロール合成がeLFにより回復したことが示唆された。以上より、eLFはHFCDによるコレステロール代謝異常からホメオスタシスを防御することにより、高コレステロール血症と粥状動脈硬化症を防ぐことが示唆された。

In recent years, animals are increasingly being transported by vehicle. The mitigation of stress animals aresubjected to while being transported by vehicle is desirable from the perspective of animal welfare. Weinvestigated the potential of acupuncture, which has fewer side effects than drug therapy, as a means fortreating canine transportation stress. Eleven dogs which had exhibited motion sickness symptoms duringtransportation (vomiting, salivation, and a reduced amount of spontaneous activity) underwent an easy-to-applyacupuncture treatment. Short, ultra-thin circular transdermal needles were applied to locations correspondingto acupoints on the apical area of both ears, which are identified as Erjian Points 1 to 3 (“Jisen” in Japanese) intraditional oriental medicine. Motion sickness symptoms were suppressed in the treated dogs the next timethey were transported. We consider acupuncture to be a promising potential therapeutic strategy in veterinarymedicine.

Mediastinal mass and hepatomegaly were detected by computed tomography in a 10-month-old Miniature Dachshund presenting with dyspnea. The dog was diagnosed as mediastinal T-cell lymphoma with liver invasion by cytological and clonality examination. Multidrug therapy (UW-25) was not effective for the dog. As a second-line therapy, a combination treatment of toceranib phosphate and CCNU was performed based on the expression profiling of multidrug resistance genes and angiogenic genes by qRT-PCR analysis, resulting in an improved condition. However, the dog died from dyspnea at 36 days after first examination. T-cell lymphoma was confirmed by necropsy.

Eight Japanese black calves (14‐65 days old) with severe respiration insufficiency due to tracheal stenosis by rib fractures at the calving were brought to Kagoshima University Veterinary Teaching Hospital. At the first visit (NOSAI), all calves had common symptoms of coughing and wheezing sounds. All calves were towed at the time of calving. In x-ray and computed-tomography (CT) examinations, the 1st to 6th right and/or left ribs were fractured and tracheal stenosis was observed. Surgical treatment was conducted in all cases to remove the 1st to 3rd fractured ribs on one side that compressed the trachea. The respiratory condition of all calves improved from the day after surgical relocation and had completely reversed by the 14th postoperative day. From the above, CT examination was found to be useful for diagnosing tracheal stenosis due to rib fracture while removing the fractured ribs on one side is effective surgical treatment.
