微少循環系の血流の血栓形成を総合的に評価できる総合的血栓形成評価システム(T-TAS)を用いて、心房細動を模したイヌ実験モデルの急速心房ペーシング(RAP)直後の心房内と末梢領域における血液凝固を比較した。その結果、ペーシング30分後の心房血では、ARチップ(凝固系と血小板のミックスした血栓分析のためのチップ)による+10kPaまでの凝固機能評価時間(T10)と閉塞時間(OT)の両方が、ペーシング前に比べて有意に短縮した。ペーシング30分後の心房血のPLチップ(血小板血栓分析のためのチップ)によるOTは、ペーシング前に比べて有意に短縮した。一方、末梢血ではARチップとOTチップによる、T10でもOTでもRAPによって有意な変化はなかった。以上、RAP発症30分後には血液凝集の上昇が示唆されたことから、末梢血での凝集性に変化がなかったとしても、心房細動後早期に抗血栓治療を始めたほうが良いと考えられた。

マイクロミニピッグ(μMP)を用いて、肥満、血清脂質代謝、アテローム性動脈硬化、肝傷害、肝臓、全身マクロファージ活性化に対するコール酸(CA)の影響を評価した。μMPを高脂肪/高コレステロール食(HcD)群、HcDに0.7%CAを添加(HcD+CA)群に分け、8週間飼育した。飼育前後に腹部CTを行い、血糖値、血球数、血液化学、リポ蛋白質値、血圧を測定した。全器官を切除して組織学的および免疫組織化学検査を行った。mRNA発現をリアルタイムPCRで定量した。HcD+CA群はHcD群よりも内臓脂肪症、アテローム性動脈硬化の進行、高血清酸化ストレスマーカー値を示したが、血清脂質値は同じであった。肝臓では脂質蓄積が増加し、酸化ストレスマーカーの発現が高く、泡沫性クッパー細胞と星状細胞の活性化が促進し、肝細胞アポトーシスが増加し、非アルコール性脂肪性肝疾患(NAFLD)を示した。HcD+CA群では網内系、肺毛細血管、皮膚を含む種々の器官で泡沫性マクロファージの動員が観察された。CAは局所的および全身での酸化ストレスと炎症性サイトカインを発現亢進させ、種々の器官でのマクロファージを活性化させ、内臓型肥満、アテローム性動脈硬化、NAFLDを促進させた。
