Powassan virus is an emerging tick-borne flavivirus that poses a significant threat to human health. The outcome of Powassan virus infection is shaped by both viral factors and the host immune response. While this review aimed to examine the innate immune response, particularly toll-like receptor-mediated immune responses to Powassan virus, data specific to the immune response to Powassan virus remain scarce. Therefore, we focused on toll-like receptor responses to related flaviviruses to infer possible mechanisms of host response. Insights from both in vivo and in vitro studies are critical for guiding the development of effective therapeutic and preventive strategies. Currently, there are no clinically approved treatments or vaccines for Powassan virus, highlighting the urgent need for their development. We also highlight recent progress in POWV vaccine development, with an emphasis on the potential use of toll-like receptor agonists as adjuvants to enhance immunogenicity and improve vaccine efficacy.

A rapidly growing mass was observed in the left third mammary gland of a 5-and-a-half-year-old intact female northern tree shrew (Tupaia belangeri). Histopathologically, the mass was composed of numerous irregularly dilated mammary ducts with a pericanalicular or intracanalicular pattern. These mammary ducts showed a leaf-like structure due to the proliferations of surrounding spindle-shaped neoplastic cells with moderate to severe nuclear atypia. We diagnosed this case as a phyllodes tumor in a mammary gland of a northern tree shrew (Tupaia belangeri), which closely resembled those occurring in the human breast histopathologically. To the best of our knowledge, this is the first report of a phyllodes tumor in the mammary gland of a northern tree shrew (Tupaia belangeri).

Influenza A viruses (IAVs) pose a significant global threat to human health. A tightly controlled host immune response is critical to avoid any detrimental effects of IAV infection. It is critical to investigate the association between the response of Toll-like receptors (TLRs) and influenza virus. Because TLRs may act as a double-edged sword, a balanced TLR response is critical for the overall benefit of the host. Consequently, a thorough understanding of the TLR response is essential for targeting TLRs as a novel therapeutic and prophylactic intervention. To date, a limited number of studies have assessed TLR and IAV interactions. Therefore, further research on TLR interactions in IAV infection should be conducted to determine their role in host–virus interactions in disease causation or clearance of the virus. Although influenza virus vaccines are available, they have limited efficacy, which should be enhanced to improve their efficacy. In this study, we discuss the current status of our understanding of the TLR response in IAV infection and the strategies adopted by IAVs to avoid TLR-mediated immune surveillance, which may help in devising new therapeutic or preventive strategies. Furthermore, recent advances in the use of TLR agonists as vaccine adjuvants to enhance influenza vaccine efficacy are discussed.
