    The current work assessed the inclusion impacts of oilseed camelina meal (CM) as a protein source in red sea bream (Pagrus major) diets. A 45 day assessment period with 180 juveniles (6.47 ± 0.17 g) were allocated in triplicates to 4 experimental groups and fed formulated diets in which fish meal (FM) was subrogated at graded series of 0% (T1), 20.5% soybean meal (T2), 20.5% camelina meal (T3), and 33% camelina meal (T4). No noticeable alterations were observed in specific growth rate, feed intake, survival rate, hepatosomatic index, and Fulton's condition factor among the experimental groups. Fish fed the T4 diet showed considerably reduced (P < 0.05) final weight and body protein content when compared to those fed T2, T3, and basal diets. Plasma biochemical parameters show no differences (P > 0.05) in glucose, total bilirubin, total protein, total cholesterol, triglyceride, glutamyl oxaloacetic transaminase, and glutamic pyruvate transaminase. Hematocrit levels decreased noticeably (P < 0.05) in fish groups fed camelina meal in comparison to the control group. Liver and intestinal histology showed a healthy status and an increase in villus length and goblet cell number in camelina groups. Fish fed the T4 diet displayed higher expression levels (P < 0.05) of relative mRNA interleukin 1 beta (IL-1b) and tumor necrosis factor-alpha (TNF-α) in comparison to other groups. The inclusion of camelina meal (up to 20%) in red sea bream diets produced similar outcomes to specimens fed fish meal and soybean meal.

    A 60-day experiment was designed to assess the effect of different ratios of fish meal (FM): camelina meal plant protein (CM) on growth response and relative gene expression of growth-pro-moting factors, feed utilization potency, digestive enzymes activities, apparent digestibility (ADC), stress response, non-specific immunity of Pagrus major. Four isonitrogenous (490.7 g/kg of crude protein) and isolipidic (91.5 g/kg total lipid) experimental diets were formulated and designated as camelina meal (CM0), soyabean meal (SBM20.5), CM20.5, and CM33 based on protein contents. At the end of the feed trial, significantly higher (p < 0.05) weight gain, specific growth rate, and feed intake but lower feed conversion ratio were recorded in fish fed CM0, SBM20.5, and CM20.5 than fish fed CM33. The lowest growth, feed utilization, enzyme activity, and digestibility were recorded in fish fed CM33. Significantly higher pepsin, amylase, and protease activities were observed in fish fed CM0, SBM20.5, and CM20.5 diets than fish fed CM33. The highest ADC of protein was recorded in fish fed CM0, SBM20.5, and CM20.5 diets. Hematocrit levels were depressed CM33 while total serum protein, total cholesterol, triglyceride, blood urea nitrogen, total bilirubin, aspartate ami-notransferase, and alanine aminotransferase were not significantly changed by the inclusion of CM. Non-specific immune variables (lysozyme activity, peroxidase activity in serum and nitro blue te-trazolium) in fish fed CM0, SBM20.5, and CM20.5 were significantly higher than in fish fed CM33 diet. The superoxide dismutase of fish fed CM20.5 was not significantly different from CM0 and SBM20.5 (p > 0.05). Catalase and low salinity stress test show that CM0, SBM20.5, and CM20.5 were not significantly (p > 0.05) different, while CM33 was significantly lower than the rest of the diets. TBARs show that CM20.5 and CM33 diets were significantly different (p < 0.05), but CM20.5 was not significantly different from SBM20.5. Significantly higher hepatic IGF-1 and IGF-2 mRNA expression was found in fish-fed diet groups CM0, SBM20.5, and CM20.5 than fish fed CM33. The present study indicated that the addition of CM up 205 kg/kg to diet maintains growth, digestive enzymes, nutrient digestibility, immunity, stress resistance, and feed utilization efficiency of red sea bream.

    The effective use of distillery by-products of the shochu sweet potato has long been considered as a rich source of protein, fiber, amino acids, fatty acids, probiotics with a great potential as a functional feed for animals. This study applied fermented by-product of Shochu distillery (FBPSD) and salmon roe emulsion oil for rotifer enrichment to evaluate its effects on the: (i) nutritional composition of rotifers and (ii) feeding responses of Paralichthys olivaceus larvae when fed with enriched rotifers. The salmon roe oil enrichment was set as a control diet and FBPSD was supplemented at different doses (1, 5 and 10 % based on the salmon roe oil amount). The results showed that the rotifers supplemented with FBPSD at 5 and 10 % significantly enhanced protein, carbohydrate, and HUFA content. Similarly, the activity and expression profiles of targeted enzymes including trypsin, chymotrypsin, lipase and amylase effectively responded at early and middle-metamorphosis of the larvae to 5 and 10 % enrichment regiment. These two treatments were also found to be better supported in energy metabolism evidenced by Cytochrome-C-Oxidase (cox) and ATPase-pk (atp) gene expression in the larvae. Moreover, the expression levels of immune-related genes (IL-10, IL-1 beta, TNF-alpha, P2Y(6)) and antioxidant-related genes (SOD, CAT) were significantly improved at 5 and 10 % of FBPSD. Cessation of feeding and delay in growth during the metamorphic process was also molecularly demonstrated. Larvae suffered low survival (24.5-25.3 %) in control and 1 % supplementation of FBPSD, but significantly improved at 5 and 10 % (36.4-38.7%). However, the growth performance of larval fish was not remarkable after 40 days of culture. The results of this study revealed that FBPSD could be considered as a potential supplementary enrichment for live feed in fish larviculture, especially at 5 and 10 % rotifer enrichment dosage.

    The euryhaline copepod <italic>Pseudodiaptomus inopinus</italic> play important roles in coastal waters as vectors of docosahexanoic (DHA) and eicosapentaenoic (EPA) acids for larval fish. While DHA and EPA in polar lipids (PLs) are more effective for fish larval development than non-polar lipid forms (NLs), there is little knowledge how much these lipids are accumulated in copepods from microalgae and are effective for early development of fish larvae. We report PLs fatty acid profiles of <italic>P. inopinus</italic> fed DHA-poor microalgae and evaluate its significance as a food source for larvae development of <italic>Pagrus major</italic>, compared with DHA-enriched rotifers. Copepods and rotifers were fed a mixed diet of three algal species (<italic>Phaeodactylum tricornutum</italic>, <italic>Tisochrysis lutea</italic>, and <italic>Pavlova lutheri</italic>), in addition of DHA-supplemented Super Fresh Chlorella (SFC) for rotifers. Compared with SFC, the algal mixture had higher EPA but lower DHA. Copepods had higher DHA and EPA in total lipids than rotifers fed each diet. Copepod PLs were specifically enriched with DHA and their contents were higher than both rotifers. On the other hand, PLs EPA contents were comparable between preys, indicating that copepods selectively fortified the PLs. Fish culture experiment showed that larvae fed copepods had higher growth than those fed SFC-enriched rotifers. Principal component analysis for each organism fatty acid composition emphasized trophic modification of DHA by copepods toward larval fish. This study highlighted that <italic>P. inopinus</italic> contribute to enhanced growth of coastal larval fish by efficiently transferring DHA via copepod fatty acid metabolism.

    A good understanding of the morphological and functional changes during early larval fish ontogeny is essential for the enhancement of larviculture techniques. This study applied an integrative approach to describe the ontogenetic development of the Japanese flounder (Paralichthys olivaceus) emphasizing the changes in histology and related functions of its digestive system. Changes in histological structures of the digestive tract, expression and activity of digestive enzymes (trypsin, chymotrypsin, lipase, pepsin, amylase), expression of energy metabolism (ATP synthase (atp), cytochrome-c-oxidase (cox), regulatory factors (neuropeptide Y (npy), cholecystokinin (cck)) and growth (insulin like growth factor 1 (igf1), growth hormone (GH)) were investigated and linked to larval biometrics. The larvae absorbed nutrients from its yolk upon hatching until mouth opening (3 DAH), before beginning its exogenous nutritional phase. The digestive tract differentiations were histologically observed at 5 DAH. The results showed that the targeted enzymes were genetically programmed and activated from hatching, except pepsin which was detected at 25 DAH coinciding with visible development of the gastric glands. The larval stages (3-5 DAH) were marked with high mortality at exogenous feeding transition and also at the onset of metamorphosis (20-30 DAH). Energy metabolism, growth and peptide regulatory factor related genes were transcribed and translated at hatching to assist upon the onset of exogenous feeding. Less feeding and growth delay were noticed between 20 and 30 DAH. The digestive system was fully developed by 35 DAH and larvae transited to settlement life.

    Kawakawa tuna (Euthynnus affinis) is a novel target species for aquaculture in Japan and currently there is very limited knowledge on its early ontogenetic development in captivity. The ontogeny of E. affinis larvae from hatching to 20 days after hatching (DAH) was illustrated in the present study through an integrated approach of histology, biochemistry and molecular approaches. Larvae were fed enriched L-type rotifer Brachionus plicatilis species complex from 2 to 10 DAH, followed by enriched Artemia nauplii. Target digestive enzymes; trypsin, pepsin, lipase and amylase were assessed in order to evaluate nutritional condition. The larvae began exogenous feeding from 2 DAH, coinciding with the opening of the oral capacity and anus. The yolk sac was rapidly consumed and completely resorbed at 3 DAH, while the oil globule disappeared by 5 DAH. Enzymatic activity and expression of the target enzymes were detectable at hatching, except pepsin. With the development of the gastric glands, pepsin activity was then detected at 10 DAH, which was followed by rapid increase in pepsin activity at 15 DAH. The larvae showed rapid growth, however, high mortality also occurred at 6-10 DAH which has been highlighted as a critical stage in the larviculture of E. affinis. From 15 to 20 DAH, the histological structures of the digestive tract were completely segmented with the digestive tract resembling that of its juvenile stages. The activity of trypsin, pepsin, and lipase tended to increase sharply, while amylase dramatically dropped, possibly indicating a change in dietary preference. Findings from the present study provides a preliminary insight into the early development of the digestive system and associated digestive enzymes of E. affinis larvae and establishes an initial step towards the rearing and feeding protocol optimization of this novel target species.

    We propose a new analytical method for determining the response threshold in electroretinogram (ERG) in which the wave shows a biphasic slow dc-potential shift. This method uses the recorded wave to the highest intensity stimuli in each wavelength tested as a template wave f(t), and it was compared with other recorded waves obtained under lower intensities g(t). Our test recordings in medaka Oryzias latipes were analogous between the template and the compared waveforms, although there were differences in amplitude and time lag (τ, peak time difference) which occurred as a result of the difference in stimulus intensity. Cross-correlation analysis was applied. Based on the obtained cross-correlation function Cfg(τ) in each comparison, τ was determined as the time lag at which the cross-correlation coefficient Rfg(τ) showed the maximum value. Determined thresholds that were based on both the experimenter's visual inspection and this new method agreed well when the adoption condition was set to satisfy R(τ) ≥ 0.7 and τ ≤ 150 ms in scotopic or τ ≤ 120 ms in photopic conditions. We concluded that this "template wave matching method" is a quick and reliable objective assessment that can be used to determine the threshold. This study analyzed ERG recordings in response to 6 kinds of wavelength light stimuli (380 nm to 620 nm) at different photon flux densities. We report the threshold levels and relative spectral sensitivities in scotopic and photopic vision of medaka.

    <p><i>Brachionus plicatilis </i>複合種 SS および L 型に及ぼす3 種の栄養強化剤(DHA 強化クロレラ,冷凍ナンノクロロプシスおよび乳化筋子油)の餌料効果を比べた(対照区:濃縮淡水クロレラ)。両型ワムシの増殖速度と脂肪酸含量は試験区間で変化した。 非極性脂質と極性脂質の含量はワムシ型間で変化した。可溶性タンパク質は両型で DHA 強化クロレラおよび生クロレラ給餌で多かった。冷凍ナンノクロロプシスを除いた全ての試験区で,SS 型の可溶性タンパク質含量は L 型よりも高かった。 ワムシ型に応じて,培養方法,強化剤の投与量および脂肪酸要求量を考慮する必要がある。</p>

    The feeding effects of freshwater cladoceran Moina macrocopa to the larvae and juveniles of red sea bream were investigated. M. macrocopa was mass-cultured with freshwater Chlorella as feed and was enriched nutritionally with a commercial enrichment diet. Because M. macrocopa die in seawater within a few minutes, the cladocerans were frozen after nutritional enrichment. The survival rates, growth rates and stress tolerances of fish fed with enriched M. macrocopa were compared with those fed with enriched Artemia and with zooplankton harvested from open seawater. The fatty acid composition of each type of zooplankton was analyzed. Mass-cultured M. macrocopa could be enriched with a commercial enrichment diet. The DHA/EPA ratio of M. macrocopa was inferior to that of enriched Artemia nauplii and wild zooplankton. Although the growth and stress tolerance of fish fed on frozen M. macrocopa were inferior to those fed with enriched Artemia nauplii and wild zooplankton, their survival rate was superior. It was concluded that frozen and enriched M. macrocopa could be fed to red sea bream larvae and juveniles. However, it is necessary to improve its fatty acid composition, particularly the DHA content.

    In order to improve the survival and growth of aquacultured finfish larvae, nutritional enrichment of rotifers is necessary. Although the addition of those fatty acids to rotifer was conducted in vitro, the enrichment of DHA or EPA to phospholipid was suggested recently, as well as the optimum ratio among DHA, EPA and arachidonic acid. On the other hand, other nutrients, e.g., taurine, vitamin A and some minerals have also been suggested to enrich rotifers. These means that not only specific nutrients but also the balance among nutrients are important to enrich rotifers nutritionally, and improve the performance of cultured finfishes. Moreover, the influence by rotifer culture method and protocol, and the development of enrichment method should be taken into consideration for improving the efficiency of enrichment. Future studies should investigate whether rotifers can acquire all these nutrients from the balanced nutrition.
