    <p> <i>Bacteroides fragilis</i> is a member of normal intestinal flora and is known as an opportunistic infectious bacterium causing intraperitoneal abscess or septicemia. It has been shown that the bacterium activates the intestinal innate immune system via Toll-like receptor 2 (TLR2), and the relation with intestinal disease has been drawing attention.</p><p> The cell surface of this bacterium is composed of capsular polysaccharide (CPS), lipopolysaccharide (LPS) and lipoprotein (LP). Capsular polysaccharide A (PSA), a type of CPS, is a zwitterionic polysaccharide and is known to induce regulatory T cells in the intestinal tract and inhibit enteritis. In recent years, it has been reported that PSA activates TLR2 which is an innate immune receptor. However, the structure of PSA is largely different from the general recognition structure of TLR2, and the possibility of contamination could be considered. In this study, we separated the PSA fraction from <i>B</i>. <i>fragilis</i> and examined the substances involved in TLR2 activation.</p><p> <i>B</i>. <i>fragilis</i> cells were subjected to hot water-phenol extraction and followed by hydrophobic chromatography separation to obtain a PSA fraction having the ability to activate TLR2. The fraction was further subjected to SDS-PAGE separation based on their molecular weight. TLR2-stimulating activity was scarcely observed in the high molecular mass area where PSA was present, and high activities were observed in the low molecular mass area. The active substances were found to be proteins because they were digested with proteolytic enzymes. These results indicate that contaminating proteins are substances responsible for TLR2 activation in the PSA fraction derived from <i>B</i>. <i>fragilis</i>.</p>

    Bacteroides fragilis is a member of the normal intestinal flora and is involved in host immunostimulation via TLR2. On the bacterial cell surface, glycoconjugates, such as LPS and capsular polysaccharide A (PSA), have been reported to participate in host immunostimulation via TLR2. Previously, we identified a TLR2-stimulating lipoprotein in B. fragilis cells. In this study, we demonstrated that TLR2-stimulating principal molecules in glycoconjugate fractions prepared from B. fragilis are contaminating proteinous molecules, which may also be lipoproteins. The glycoconjugate fractions were prepared by phenol-hot water extraction of B. fragilis wild type and PSA-deficient strains, followed by hydrophobic interaction chromatography. TLR2-stimilating activities of the fractions were not affected by PSA deficiency. By in-gel TLR2-stimulation assay, molecules in high-molecular-mass area, where capsular polysaccharides were migrated, were found not to stimulate TLR2, but those in the range of 15-40kDa were active. Further, proteinase K could digest the latter molecules and the TLR2-stimulating activities were migrated to the area of below 15kDa. These results support that proteinous molecules, which are estimated to be lipoproteins, are responsible for almost all TLR2-stimulating activity in the glycoconjugate fractions prepared from B. fragilis.

    ファージ(バクテリオファージ)は細菌に感染して増殖するウイルスである。ファージを担体としたワクチン開発は古くから研究されているが、ファージは哺乳動物には感染することがなくヒトへの投与実験において安全であることが示されており、アジュバントを用いることなく体液性免疫および細胞性免疫の両者を誘導できる、非侵襲的な投与も可能、物理化学的に過酷な条件でも安定であるなどの利点により、ファージを担体としたワクチン開発が注目されつつある。ファージワクチンには、(1)ワクチン抗原をファージ表面に提示させたファージ粒子をワクチンとして利用する試みと、(2)ファージをDNAワクチンの運搬体として利用する試みがある。本稿では、ファージディスプレイ法を利用した繊維状ファージワクチン研究の話題を中心に、この領域の概要を述べる。(著者抄録)

    2000年頃、網膜にメラノプシン細胞と呼ばれる新たな光受容器が発見された。メラノプシン細胞は概日リズムや瞳孔の対光反射、明るさの知覚等、様々な影響を生体に与えることが報告されている。研究代表者は先行研究においてメラノプシン細胞のみを選択的に刺激することが可能な刺激装置を開発し、メラノプシン細胞の生体への機能解明に取り組んでいる。本研究課題では、メラノプシン細胞の生体への影響を実験心理学、生物学、工学的手法を用いて包括的に解明することを目指す。2020年度は、辻村らが開発した多原色光源刺激装置を用いて、メラノプシン細胞のみを選択的に刺激し、明るさ知覚への寄与に関する実験を立ち上げた。従来の刺激条件よりも自然光に近いスペクトラムを用いた。このことにより、より自然環境に近い光環境で実験を進めることが可能となることが期待される。この実験環境によって、心理物理学実験を実施し、基礎的なデータを取得した。国立台湾大学とメラノプシン細胞の視聴覚統合過程への影響に関する論文、および高知工科大学とメラノプシン細胞の明るさ知覚への寄与についての論文をまとめた。
    さらに国立台湾大学との共同研究を進め、メラノプシン細胞の明るさコントラスト感度への寄与について、詳細にデータを解析し現在論文の執筆中である。工学的研究ではこれまでに得られた知見をもとに新たな多原色光源刺激装置を試作した。従来の装置と比較して、光源を安定化させることにより、高精度の刺激の提示が可能になることが期待される。2021年度には装置の性能をより詳細に検証し、引き続き開発を実施する予定である。

    本研究では、細菌に感染するバクテリオファージ(ファージ)の医療応用を目的として、ファージが生体に与える影響について解析するとともに、ファージワクチンやファージ療法への応用について検討を行っている。本年度は繊維状の形態のM13ファージに加えて、緑膿菌を溶菌するファージおよび黄色ブドウ球菌を溶菌するファージを取得しマウスへの投与実験を行った。外来のペプチド分子を提示させたM13ファージを用いてTLR9欠損マウスに投与し抗体応答の解析を行った結果、野性型のC57BL/6マウスで誘導されるIgGサブクラスに加えて、C57BL/6マウスではほとんど検出されないIgG1クラスの抗体が、TLR9欠損マウスにおいて一次応答の段階から強く増強された。一方、MyD88欠損マウスにおいては抗体応答が惹起されなかった。これらの結果は、野性型M13ファージを免疫した以前の研究と同様の結果であり、TLR9がIgG1サブクラス応答にどのように寄与するか、現在も引き続き検討を進めている。また、緑膿菌、黄色ブドウ球菌などの細菌株を用いて自然環境中から単離したファージクローンのマウスへの投与実験においては、M13ファージと同様に、ファージ投与による顕著な体温変化および体重変化は認められなかった。この結果より、環境中から単離したファージの哺乳動物への投与は安全であることが示唆された。現在、溶菌活性を有するファージクローンに対する免疫応答についても解析を進めている。

    In this study, we conducted research on the practical application of phage therapy using bacteriophages that infect and kill only bacteria against drug-resistant bacterial infections. By analyzing in the mechanism of phage resistance in bacteria, this study clarified the mechanism of phage resistance and laid the foundation for designing strategic phage cocktail therapy. In addition, for the first time in Japan, a successful clinical trial of phage therapy was conducted on clinical cases in dogs and cats. We believe that the details of the phage resistance of bacteria in this clinical trial and the design of phage cocktails to deal with it will be valuable knowledge for future clinical applications in humans, and we plan to report them widely with the publication of a paper.

    There are photoreceptors in the retina, cone and rod cells. In addition, the third type of photoreceptor was discovered around 2000 which is called melanopsin cells. In this study, we aimed to clarify how the non-imaging pathway originating from melanopsin cells processes and sends light signals in the brain by using a multi-primary stimulation system that can independently control the amount of stimulation of melanopsin cells and the other photoreceptors. We have shown that selective stimulation of melanopsin cells affects the perception of time. Furthermore, we quantified the effect on the perception of brightness from melanopsin cells.

    A novel LRR antibody was designed based on LRR scaffold of human TLR3 protein. The designed antigen binding sites were constructed of several repetitive LRR modules. In this study, a novel library construction method for repetitive protein was designed. LRR antibody library was constructed and evaluated its properties. In this method, several randomized LRR modules were recombined to the phagemid vector in one step. This is easy method for construct repetitive protein library with retaining its diversity.
