Citrus fruit production in Japan currently stands at approximately 1.022 million tons, making it the number one fruit species cultivated domestically. Shiikuwasha (Citrus depressa Hayata) and Tachibana [C. tachibana (Makino) Tanaka] have grown wild in Japan from ancient times, so the citrus fruits currently on the market have either been introduced from overseas or developed through natural or artificial hybridization among the introduced species/cultivars and the two wild species. In Japan, after the Meiji Restoration (1868), cultivation of Satsuma mandarin began nationwide. From that time onwards, sweet orange, ponkan, and lemon were also introduced from overseas. Satsuma mandarin production increased from the 1950s, but gradually declined in the late 1970s. A breeding program was launched by a national research institute in 1937, and the first hybrid cultivar, ‘Kiyomi’, was registered in 1979, which led to the development of many excellent cultivars derived from ‘Kiyomi’, including ‘Shiranuhi’, ‘Harumi’, ‘Setoka’, and ‘Beni Madonna’. Meanwhile, research on citrus cultivation, storage, and sorting systems has progressed steadily. Japan has developed unique cultivation techniques such as mulch cultivation systems and plastic house cultivation for producing high-quality citrus. Regarding storage techniques, development is also underway on low-temperature storage with cool air and high humidity, and decay prevention techniques using ozone and UV irradiation. In addition to the conventional near-infrared method of determining sugar content and acidity, non-destructive sorting systems also use artificial intelligence (AI) technology, which has still room for further development over the next ten years. In the future, the citrus industry will also be faced with the urgent task of achieving labor-saving, low-cost cultivation, stable production, and adding high value to the products, as well as securing successors with a view to corporate management. We describe here cultural environment, cultivars and rootstocks, breeding program, cultural practice, pest and diseases, and marketing system of citrus fruit to overview the current status of citrus industry in Japan for better future.

The long-term preservation of pollen is imperative for the conservation of genetic resources and cross-breeding. In the present study, we demonstrated that preserved pollen of the Japanese pear (Pyrus pyrifolia (Burm. f.) Nakai) retains viability for a minimum of 50 years. In 1973, fresh pollen from ‘Chojuro’ was collected and immediately subjected to freeze-drying for 15 minutes at pressures ranging from 3.3 to 13 Pa. The pollen was then sealed in ampules under a vacuum and preserved at −20°C for 40 years followed by −30°C for 10 years. In 2013 and 2023, the viability of this pollen was investigated. The rate of pollen germination showed no significant differences between the preserved and fresh pollen. In 2013, flowers of ‘Kosui’ and ‘Niitaka’ were pollinated with pollen preserved for 40 years as well as fresh pollen. The growth of the pollen tubes in the pistils of preserved pollen was normal. Pollination with preserved pollen resulted in 77.1 and 86.7% fruit set, yielding 3.2 and 8.3 perfect seeds per fruit in ‘Kosui’ and ‘Niitaka’, respectively. In 2023, the fruit set and perfect seeds per fruit of ‘Kosui’ pollinated with 50-year-preserved pollen were 66.7% and 3.5, respectively. There were no significant differences between preserved and fresh either year for either trait. The results of the present study demonstrate that the viability of pear pollen can be sustained for a minimum of 50 years when preserved in a freeze-dried state at temperatures ranging from −20 to −30°C.

A chromosome preparation method using young leaves of acerola (Malpighia glabra) was developed. The young leaves were cut into approximately 2-mm2 for enzymatic maceration and air-drying (EMA). For EMA, an enzyme mixture containing 2% Cellulase Onozuka RS, 1.5% Macerozyme R200 (Yakult), 0.3% Pectolyase Y-23 (Seishin Pharmaceutical Co., Ltd.), and 1 mM EDTA, pH 4.2, at 37°C for 30 min was optimal for chromosome preparation since good preparations, with all 20 chromosomes relatively extended and well-spread without cytoplasm, were observed. There were 11.2 preparations in large leaves (8 to 10 mm long and 3 to 4 mm wide) and 4.2 preparations in small leaves (6 to 8 mm long and 1 to 2 mm wide). Chromomycin A3 (CMA)-positive (+) bands were noted in the telomeric positions of eight chromosomes. 4′-6-diamidino-2-phenylindole (DAPI)-negative bands (−) corresponded to CMA+ bands. The numbers and positions of CMA+ bands were the same in the two cultivars examined: ‘Florida Sweet’ and ‘Sanmi-kei (Hosoba).’ The methods developed in the present study are promising for further cytogenetic studies in acerola.
