    下垂体アデニル酸シクラーゼ活性化ポリペプチド(PACAP)は視床下部に高発現する神経ペプチドで、摂食制御に重要な役割を担っているが、その摂食制御メカニズムは完全に解明されていない。本研究では、PACAPノックアウト(KO)マウスと視床下部腹内側核PACAP過剰発現マウスを用いて、特に視床下部腹内側核に発現するPACAPが、明期には摂食抑制的に、暗期には摂食亢進的に働く可能性を明らかにした。(著者抄録)

    アストロサイト-ニューロン乳酸シャトル(ANLS)は、アストロサイトを含むグリア細胞が産生した乳酸をニューロンが利用することを仮定したモデルである。ANLSは近年、学習・記憶、薬物依存などに重要な役割を果たし、細胞レベルにおいても、ニューロンを直接活性化したり、シナプス促通を惹起したりすることが報告されている。しかしながら、どのようなシグナルがアストロサイトに作用し、ANLSを活性化するか明らかではなかった。一方、神経ペプチド、下垂体アデニル酸シクラーゼ活性化ポリペプチド(PACAP)とその受容体は、中枢神経系に広く分布する。我々は前脳由来培養アストロサイトにおいて、PACAPは非常に低い濃度からANLSを活性化する可能性を見出したことから、内因性のANLS活性化因子の候補としてPACAPを捉え、恐怖記憶、脊髄痛覚伝達に対する関与を検討した。その結果、PACAPは少なくとも前脳と脊髄のアストロサイトのPACAP/PAC1シグナルの下流においてPKCを介したANLSの活性化を惹起し、恐怖記憶および脊髄痛覚伝達に関与することが明らかとなった。以上の結果から、PACAPは内因性のANLS活性化因子である可能性が示された。(著者抄録)
