    <p>Seed development is an essential phenomenon for all sexual propagative plant species. The functional allele at <i>SEED DEVELOPMENT 1</i> (<i>SDV1</i>) or <i>SEED DEVELOPMENT 2</i> (<i>SDV2</i>) loci is essential for seed development for <i>Oryza sativa</i> and <i>Oryza meridionalis</i>. In the present study, we performed fine mapping of <i>SDV1</i>, narrowing down the area of interest to 333kb on chromosome 6. Haplotype analysis around the <i>SDV1</i> locus of <i>O. ‍meridionalis</i> accessions indicated that they shared the DNA polymorphism, suggesting that they have a common abortive allele at the <i>SDV1</i> locus. Linkage analysis of the candidate <i>SDV2</i> gene showed that it was located on chromosome 4. The candidate <i>SDV2</i> was confirmed using a population in which both the <i>SDV1</i> and <i>SDV2</i> genes were segregating. The chromosomal region covering the <i>SDV1</i> gene was predicted to contain 30 protein-coding genes in <i>O. ‍sativa</i>. Five of these genes have conserved DNA sequences in the chromosomal region of the <i>SDV2</i> gene on chromosome 4, and not on chromosome 6, of <i>O. ‍meridionalis</i>. These results suggest that these five genes could be candidates for <i>SDV1</i>, and that their orthologous genes located on chromosome 4 of <i>O. ‍meridionalis</i> could be candidates for <i>SDV2</i>.</p>

    Bacterial blight is an important rice disease caused by bacteria named Xanthomonas oryzae pv. oryzae (Xoo). XM5 is an Xoo resistant mutant line with the genetic background of IR24, an Indica Xoo susceptible cultivar, induced by a chemical mutagen N-methyl-N-nitrosourea (MNU). XM5 carries a recessive Xoo resistant gene, xa19. Trisomic analysis was conducted using the cross between XM5 and the trisomic series under the genetic background of IR24, showing that xa19 was located on chromosome 7. The approximate chromosomal location was found using 37 surely resistant plants in the F2 population from XM5 × Kinmaze, which was susceptible to most Japanese Xoo races. The IAS44 line carries a Japonica cultivar Asominori chromosomal segment covering the xa19 locus under the IR24 genetic background. Linkage analysis using the F2 population from the cross between XM5 and IAS44 revealed that xa19 was located within the 0.8 cM region between RM8262 and RM6728. xa19 is not allelic to the known Xoo resistant genes. However, its location suggests that it might be allelic to a lesion-mimic mutant gene spl5, some alleles of which are resistant to several Xoo races. Together with xa20 and xa42, three Xoo resistant genes were induced from IR24 by MNU. The significance of chemical mutagen as a source of Xoo resistance was discussed.

    <p>A shortage of seedlings of <i>Peucedanum japonicum</i> Thunb. var. <i>latifolium</i> M.Hotta et Shiuchi in Minamisatsuma city of Kagoshima prefecture has become a serious problem recently because <i>P. japonicum</i> plants are popular as a healthy vegetable in islands of Kagoshima and Okinawa prefecture. In our interview with farmers and research, the reason for it was a low germination percentage of <i>P. japonicum</i> var. <i>latifolium</i>. The objective of our research was to clarify the temperature which increased germination percentages. One hundred seeds were put in 9 cm-diameter petri dishes with 10 mL distilled water, dishes were tested in 24hrs dark conditions in incubators. We also compared the germination of seeds with decortication and control treatments. In 24hrs constant temperature treatments (20, 25, and 30℃), about 40% of germination percentages were observed at 25℃ with decortication, and lower germination percentages were at 20℃. No germination was observed at 30℃. In 12/12hrs alternative temperature treatments(10/25℃, 20/25℃, 20/30℃, and 25/30℃), seeds could not germinate at 10/25℃, 20/30℃ and 25/30℃, and control at 20/25℃ treatments showed about 10% germination percentages, while decortication treatments at 15/20℃, 15/25℃ showed more than 20%, and more than 40% at 20/25℃, especially. Therefore, it was estimated that daily changes of temperature could affect germination severely, and for the increase of germination percentages, the temperature has to be adjusted 20/25℃ and it will be better with decortication.</p>

    Various kinds of reproductive barriers have been reported in intraspecific and interspecific crosses between the AA genome Oryza species, to which Asian rice (O. sativa) and African rice (O. glaberrima) belong. A hybrid seed sterility phenomenon was found in the progeny of the cross between O. sativa and O. meridionalis, which is found in Northern Australia and Indonesia and has diverged from the other AA genome species. This phenomenon could be explained by an egg-killer model. Linkage analysis using DNA markers showed that the causal gene was located on the distal end of chromosome 1. Because no known egg-killer gene was located in that chromosomal region, this gene was named HYBRID SPIKELET STERILITY 57 (abbreviated form, S57). In heterozygotes, the eggs carrying the sativa allele are killed, causing semi-sterility. This killer system works incompletely: some eggs carrying the sativa allele survive and can be fertilized. The distribution of alleles in wild populations of O. meridionalis was discussed from the perspective of genetic differentiation of populations.

    <p>Bacterial blight caused by <i>Xanthomonas oryzae</i> pv. <i>oryzae</i> (<i>Xoo</i>) is an important disease constraining rice (<i>Oryza sativa</i> L.) production worldwide. The XM6 line was induced by N-methyl-N-nitrosourea from IR24, an Indica cultivar that is susceptible to Philippine and Japanese <i>Xoo</i> races. XM6 was confirmed to carry a recessive gene named <i>xa20</i>, resistant to six Philippine and five Japanese <i>Xoo</i> races. The chromosomal gene location was found using 10 plants with the shortest lesion length in an F<sub>2</sub> population consisting of 298 plants from a susceptible Japonica variety Koshihikari × XM6. Analysis using PCR-based DNA markers covering the whole rice genome indicated the gene as located on the distal region of the long arm of chromosome 3. The IKC3 line carries IR24 genetic background with Koshihikari fragment on chromosome 3 where a resistance gene was thought to be located. The F<sub>2</sub> population from IKC3 × XM6 clearly showed a bimodal distribution separating resistant and susceptible plants. Further linkage analysis conducted using this F<sub>2</sub> population revealed that <i>xa20</i> is located within the 0.8 cM region flanked by DNA markers KIC3-33.88 (33.0 Mb) and KIC3-34.06 (33.2 Mb). This study yields important findings for resistance breeding and for the genetic mechanism of <i>Xoo</i> resistance.</p>

    <p>To elucidate the diversity and evolution of the <i>Si7PPO</i> gene that controls phenol color reaction (Phr) in foxtail millet, <i>Setaria italica</i>, we analyzed sequence polymorphisms of the <i>Si7PPO</i> gene in 39 accessions consisting of foxtail millet landraces (32 accessions) and their wild ancestor ssp. <i>viridis</i> (seven accessions) collected from various regions in Europe and Asia. The accessions included wild type (positive Phr) and three different types of loss-of-function phenotype (negative Phr), "stop codon type", "TE1-insertion type" and "6-bp duplication type", found in our previous study. We constructed a phylogenetic tree of the gene and found that accessions with positive Phr showed higher genetic diversity at the nucleotide sequence level. We also found that the three different loss-of-function types formed different clusters, suggesting that landraces with negative Phr have multiple origins from three different lineages including both landrace and ssp. <i>viridis</i> accessions with positive Phr.</p>

    We genotyped Setaria italica Dehydration-responsive element-binding 2 (SiDREB2) gene, which had been reported to be associated with dehydration stress tolerance, in 588 accessions of foxtail millet from various parts of Eurasia and other regions by a dCAPS marker. Of these, 480 accessions were genotyped in ribosomal DNA (rDNA), polyphenol oxidase (PPO) gene and Heading date 1 (HD1) gene in our previous studies, and 108 accessions from India were newly used in this study and genotyped in these genes in addition to the SiDREB2 gene. We compared the geographical distribution of the SiDREB2 genotypes with that of these three genes. Accessions from countries in South Asia including India and Sri Lanka were the most variable in the SiDREB2 gene followed by Korean accessions and Japanese accessions, suggesting that Indian accessions and East Asian accessions are useful as genetic resources for dehydration stress tolerance. This study also showed that although Indian accessions are not so diverse in rDNA and in transposon display markers previously studied, they are diverse in adaptive genes such as SiDREB2, PPO and HD1 genes.

    Improvement of resistance against rice bacterial blight (BB) disease is an important breeding strategy in breeding programs across the world, especially in Africa and southern Asia where BB is more prevalent. This report describes a high-resolution map and characterization of xa42 at XA42 locus, a rice BB resistance gene in XM14, a mutant line originating from IR24. The candidate gene region was narrowed down from 582 kb, which had been obtained in our previous study, to 57 kb. XM14 shows brown spots in its leaves like lesion mimic mutants. This line also shows a shorter stature than the original cultivar IR24. In XA42 gene segregating populations, homozygotes of xa42 allele were consistently resistant to the six Japanese Xanthomonas oryzae pv. oryzae races used for this study. They also showed brown spots and markedly short stature compared with the other genotypes, suggesting that xa42 gene exhibits pleiotropic effects.

    鹿児島大学農学部付属農場の1枚の水田(土壌総水銀濃度:0.2 mg/kg)を利用し、遺伝子的に大きく異なるコメの品種(WRC系統59品種)を育て、各品種の玄米に含まれる水銀濃度を測定した。この結果、最高 7.1 ng/g 最低1.7 ng/g (平均値 3.2 ng/g, 中央値 3.0 ng/g, n = 59)となり、同一土壌で育成した稲であっても品種によって含まれる水銀濃度が異なることを確認した。一方、水銀汚染土(水田土壌に2015年に10 mg/kgとなるように水銀イオンを添加し、毎年稲の栽培に利用している土壌、土壌総水銀濃度:8 mg/kg)を利用し、予備的に選ばれた5品種についてポット栽培を行った。2週間に一度、土壌を採取し、土壌のメチル水銀濃度の変化および稲に含まれる水銀量を測定した。玄米の水銀濃度は、日本晴(WRC1)で0.25 mg/kg となり、含まれる水銀のほとんどがメチル水銀であることを確認した。土壌のメチル水銀濃度が、湛水から1ヵ月程度で最大(約0.1 mg/kg)となり、徐々に低下していくこと、一方、稲は品種により出穂までの育成期間やバイオマスが大きく異なることから、土壌メチル水銀の影響を受けやすい、受けにくいの差が生じ、玄米の水銀濃度に品種差が生じている可能性も示唆された。
    土壌におけるメチル水銀生成と、存在する無機水銀形態の関係を明らかにするため、土壌に含まれる無機水銀形態を簡易に分析する手法について検討を進めた。様々な水銀汚染地域で得た土壌試料を利用し、加熱温度を段階的に変化させつつ、気化する水銀量を評価することで、存在する水銀形態の判別を試みた。試料によっては特徴的な結果が得られたが、気化温度と水銀形態の関係は明確には確認できなかった。今後、試料数を増やしつつ、存在形態別分析法として一般的に用いられている逐次抽出法と比較していく予定である。

    白葉枯病はイネの最重要病害の一つである.インド型イネ品種IR24はフィリピンの白葉枯病菌6レースと日本の6レースすべてに罹病性であるが,人為突然変異処理によりそれらすべてに抵抗性を示す系統XM5, XM6, XM14が得られている.各系統は劣性の1抵抗性遺伝子を持ち,それぞれxa19, xa20, xa42と命名された. 2021年度の実績を以下に示す.
    (1)イネ品種 日本晴に野生型のXa42遺伝子で形質転換した個体とXM14を交配し,得られたF1個体を自殖させてF2となる種子を得た.また,日本晴にゲノム編集を行い、野生型Xa42遺伝子が変異型遺伝子ホモ接合に変換された個体を複数得た。
    (2)XM14の擬似病斑の組織構造および発達経過を観察した.走査電子顕微鏡で観察したところ表面における異常は観察できなかった.病斑を表皮組織よりも深く分析できるX線解析顕微鏡で観察したところK,Si, CaおよびClについて分布に粗密が見られる試料が見られた.
    (3)通常栽培で播種後25日のXM14, IR24ではROSとNOの両方とも、XM14の方が有意に多かった。擬似病班が出現する条件で栽培して比較したところ、両系統ともに、NO量は通常栽培より有意に高い値を示した。PR1a、PR1b遺伝子の発現は、擬似病斑が出現したXM14で非常に高かった。
    (4)xa19, xa20の候補染色体領域におけるSNPを検出できるようなDNAマーカーを設計し,白葉枯病抵抗性とDNAマーカー遺伝子型の関係を調査した. xa19, xa20, xa42遺伝子 の日本型品種台中65号背景の準同質遺伝子系統,長稈遺伝子と3抵抗性遺伝子を組み合わせたIR24背景の準同質遺伝子系統作りを進め,BC3F1となる種子を得た.

    To investigate the accumulation of methylmercury in rice grains, the uptake behavior of mercury by rice plant was examined through hydroponic experiments, which involved addition of mercury to the culture solution. In addition, the formation of methylmercury from mercury ion in waterlogged soil and the uptake of the formed methylmercury by rice plant were investigated. The results of hydroponic experiments showed that mercury ion and methylmercury, added to the culture solution, accumulated in the roots, and only methylmercury was transferred to the ears during ear growth. The concentration of methylmercury in the waterlogged soil fluctuated greatly during the rice cultivation period, with and without rice plants. Of note, methylmercury concentration increased with decreasing soil redox potential; however, with further decrease in soil redox potential, methylmercury concentration decreased.
