    Environmental DNA (eDNA) analysis with species-specific primer/probe sets is promising as a tool to quantify fish abundance and distribution. Nevertheless, several factors could reduce the accuracy of this method. Here, we aimed to analyze whether intraspecific variability and diel activity rhythm affect eDNA detection in Japanese eels (Anguilla japonica). For this purpose, we performed tank experiments focusing on two points. First, we assessed the effects of base pair sequences with probe region polymorphism on eDNA detection. Next, we evaluated the influences of diel rhythm, activity, and individual differences in eDNA release rate on eDNA concentration. We examined the base pair sequences of the probe regions of 20 individuals and found genetic mismatches in two of them. The eDNA concentration was estimated to be much lower in these variants than it was in the other individuals. We conducted a rearing experiment on four non-variant individuals to explore the influences of diel activity and inter-individual differences in eDNA detection. Nocturnal eel activity was reflected in the eDNA detection but the inter-individual differences remained large. The observed weak positive correlations between eDNA concentration and activity suggest that eDNA emission is highly dependent on basal metabolism. The present study suggests that consideration of polymorphic sites at the probe region and diel activity rhythms should improve the accuracy and precision of abundance estimation through eDNA. Such fine-tuning is applicable not only for eels but also for other fishes to be targeted by eDNA technology.

    To discuss the diversity of morphological traits and life strategies of trees, the functional relationship between leaf expansion and vessel formation must be clarified. We compared the temporal relationship among tree species with different leaf habits and vessel arrangements. Twigs, leaves, and trunk core samples were periodically acquired from 35 sample trees of nine species in a temperate forest in Japan. We quantitatively estimated leaf expansion using a nonlinear regression model and observed thin sections of twigs and trunks with a light microscope. Almost all of the first-formed vessels in twigs, which formed adjacent to the annual ring border, were lignified with a leaf area between 0% and 70% of the maximum in all species. The first-formed vessels in trunks lignified between 0% and 95% of the maximum leaf area in ring-porous deciduous Quercus serrata and ring-(radial-)porous evergreen Castanopsis cuspidate. Their lignification occurred earlier than in diffuse-porous deciduous Liquidambar styraciflua, diffuse-porous evergreen Cinnamomum camphora and Symplocos prunifolia, and radial-porous evergreen Quercus glauca and Quercus myrsinifolia. The timing varied in semi-ring-porous deciduous Acanthopanax sciadophylloides and diffuse-porous evergreen Ilex pedunculosa. The observed differences in the timing of vessel formation after leaf appearance were reflected in their differing vessel porosities and were connected to the different life strategies among tree species.

    Research on the functional relationship between leaves and conductive tissues of trees is very important, as it can lead to new knowledge on tree growth and life. Although previous studies have examined leaf expansion qualitatively, quantitative estimation of leaf expansion plays an important role in analysis of physiological leaf phenology. So far, three curves, namely the Mitscherlich, logistic, and Gompertz curves have been applied to best fit the time course of leaf expansion, while Richards’ growth function has not been previously evaluated. This function has five parameters (A, k, b, m, and t0), and comprises the above mentioned three types of curves which differ regarding the ‘m’ parameter. This study aimed to estimate time courses of changes in leaf area using Richards’ growth function and to develop leaf expansion model, which can be used in common without depending on a difference of tree ring porosities. Leaves were sampled periodically from ring-porous jolcham oaks, Quercus serrata, and from diffuse-porous American sweetgum trees, Liquidambar styraciflua, both of which grow in temperate forests. Richards’ growth function was able to describe seasonal changes in leaf area in both species, and an asymmetrical function of the Gompertz was suitable for them. We suggest that Richards’ growth function can be used to estimate leaf area time courses without concern about the curve type, and can further be used to calculate leaf expansion quantitatively.

    Water sampling and filtration of environmental DNA (eDNA) analysis have been performed by several different methods, and each method may yield a different species composition or eDNA concentration. Here, we investigated the eDNA of seawater samples directly collected by SCUBA to compare two widely used filtration methods: open filtration with a glass filter (GF/F) and enclosed filtration (Sterivex). We referred to biomass based on visual observation data collected simultaneously to clarify the difference between organism groups. Water samples were collected at two points in the Sea of Japan in May, September and December 2018. The respective samples were filtered through GF/F and Sterivex for eDNA extraction. We quantified the eDNA concentration of five fish and two cnidarian species by quantitative polymerase chain reaction (qPCR) using species-specific primers/probe sets. A strong correlation of eDNA concentration was obtained between GF/F and Sterivex; the intercepts and slopes of the linear regression lines were slightly different in fish and jellyfish. The amount of eDNA detected using the GF/F filtration method was higher than that detected using Sterivex when the eDNA concentration was high; the opposite trend was observed when the eDNA concentration was relatively low. The concentration of eDNA correlated with visually estimated biomass; eDNA concentration per biomass in jellyfish was approximately 700 times greater than that in fish. We conclude that GF/F provides an advantage in collecting a large amount of eDNA, whereas Sterivex offers superior eDNA sensitivity. Both filtration methods are effective in estimating the spatiotemporal biomass size of target marine species.

    In order to understand the coordination of leaf phenology and functional xylem anatomy, the timing of vessel wall lignification in twigs and stems in relation to leaf appearance was studied in nine species with different porosity patterns. Cylindrical stem cores and twigs were collected from early spring through late summer from deciduous (Quercus serrata, Liquidambar styraciflua, and Acanthopanax sciadophylloides), and evergreen (Castanopsis cuspidata; Cinnamomum camphora, Ilex pedunculosa, Symplocos prunifolia, Quercus glauca and Quercus myrsinifolia) species in a temperate forest. The first-formed twig vessels lignified at the time of leaf appearance or before in all species. The timing of stem vessel lignification in relation to leaf appearance in semi-ringporous deciduous species was overlapping with that of ring-porous deciduous species and diffuse-porous deciduous species. Evergreen species showed a great variation in the timing of stem vessel lignification, relative to leaf flushing. The main conclusions are that 1) Vessel lignification occurs much earlier in twigs than in trunks of the same trees, with hardly any overlap between the two; 2) Deciduous trees do not differ much from evergreen species, but there is a weak tendency for evergreen species to have later vessel differentiation than deciduous species; 3) The timing of vessel formation shows little relation with porosity patterns and overlaps between diffuse-porous and ring-porous species. This suggests a much greater intergradation of timing of vessel formation in species of different porosity pattern in evergreen and deciduous species than recognized in the literature.

    To elucidate the functional relationship between intra-annual variations in vessel diameter and leaf emergence pattern of ring-porous and diffuse-porous deciduous trees in temperate forests, we determined the temporal relationships between leaf phenology and vessel formation. Cylindrical stem cores were periodically collected from each of five ring- and diffuse-porous species, and the leaf and vessel formation were observed simultaneously. In the ring-porous species, vessel formation began within 2 weeks of leaf appearance, and most wide vessels were formed within 2 weeks of full leaf expansion. More of the trees with intermediate-type formed medium-sized vessels or sparse wide vessels than trees with the flush-type leaf emergence, especially between full leaf expansion and the end of shoot elongation. Narrow vessel formation began 2–8 weeks after full leaf expansion in all specimens. The transition between the pore- and non-pore zones was abrupt in the flush-type species, and gradual in the intermediate-type species. In contrast, diffuse-porous species formed vessels 0–8 weeks after full leaf expansion. Our findings suggest that ring-porous species form leaves and vessels synchronously to accommodate water transport requirements, whereas diffuse-porous species form these tissues asynchronously; thus, unlike the latter type, the former species change vessel diameters according to leaf formation.

    The goal of this study is to clarify how different aspects of plant function are coordinated developmentally for species of ring-porous versus diffuse-porous deciduous trees, comparing the timing of leaf phenology and vessel formation in twigs and stems from an ecophysiological viewpoint. Cylindrical stem cores and twigs were collected at intervals from early spring through summer from five ring-porous and five diffuse-porous species in a cool temperate forest, and leaf and vessel formation were observed simultaneously. We found that the first-formed vessels of the year were lignified in twigs around the time of leaf appearance and at or before full leaf expansion of each tree in both groups of species with flush-leaves. Vessels in stems were lignified 2 weeks before to 4 weeks after leaf appearance and before or around full leaf expansion of the tree in ring-porous species. This was significantly earlier than in diffuse-porous species, in which stem vessel lignification was 2–8 weeks after leaf appearance and at or after full leaf expansion of the tree. The timing of vessel formation in twigs compared to stems was significantly earlier in ring-porous species than in diffuse-porous species. Lignification of vessels in stems occurred within 2 weeks of lignification in the twigs of ring-porous species and 2–8 weeks after lignification in twigs of diffuse-porous species. These results indicate the order and time-lag of leaf and vessel formation. Ring-porous species showed intensive leaf/vessel production, whereas diffuse-porous species showed less intensive leaf/vesse production.
