    <p> Toll-like receptors (TLRs) are transmembrane proteins classified into pattern recognition receptors (PRRs). TLRs are found in cell surface and/or endocytic vehicles in immune cells. TLRs activate innate immunity by recognizing pathogen-associated molecular patterns (PAMPs) derived from bacteria and viruses and damage-associated molecular patterns (DAMPs) produced by damaged tissue. The activation of innate immunity induces adaptive immunity that plays an important role in the host defense against pathogenic infection. On the other hand, abnormal activation of innate immunity causes immune disorders such as autoimmune disease. Therefore, TLRs are important molecules in the immune system. So far, 10 types of TLRs (TLR1 to TLR10) in humans and 12 types of TLRs (TLR1 to TLR9, TLR11 to TLR13) in mice have been found. TLR1, TLR2, TLR4, and TLR6 are found on the cell surface, and TLR3, TLR7, TLR8, and TLR9 are found in the endosomal compartment. Each TLR recognizes specific PAMPs and DAMPs ; lipopeptides for TLR1/2 and TLR2/6 ; nucleic acid structures for TLR3, TLR7, TLR8 and TLR9 ; lipopolysaccharide (LPS) structures and heat-shock proteins for TLR4. In recent years, various ligands activating TLRs signal have also been chemically synthesized and have been utilized as effective adjuvants in antiviral and anticancer therapy. Regarding TLR7 ligand (TLR7L), conjugates with peptides, polymers, proteins, glycans, and nanoparticles have attracted much attention because they have great potencies on the humoral and cellular immune response. In this paper, we describe recent progress in the synthesis and biological property of TLR7 ligand conjugate containing our recent study.</p>

    <p> Adjuvants enhance immune system during vaccination. Among FDA-approved adjuvants, aluminum salts are most commonly used for vaccines. Although aluminum salts enhance antibody production, they show a limited effect for the cell-mediated immune response. Thus, further development of adjuvants inducing T cell mediated immuno-responses are demanded. Toll-like receptors (TLRs) are immune-related receptors that recognize specific pathogen-associated molecular patterns and play important roles in the activation of innate immunity, which is crucial to shape adaptive immunity. Studies using TLR ligands as novel adjuvants for anti-microbial and anti-cancer immunotherapies have therefore attracted much attention. Among them, a low molecular weight TLR7 ligand, Imiquimod, has been approved for clinical use, but its use is restricted only for local administration due to unwanted adverse effects. Since TLR7 is mainly located in the endosomal compartment of immune cells, efficient transport of the ligand into the cell is important for activating TLR7. Our previous work indicated that the conjugation of a low molecular weight TLR7 ligand with serum albumin and polysaccharides can greatly enhance its potency. In this study, we examined gold nanoparticles (GNPs) as carriers, as GNPs are less toxic and can immobilize multiple molecules including antigens for pathogens and tumors. Furthermore, α-mannose for targeting antigen presenting cells was also examined for the efficient delivery of GNPs. In this paper, we describe preparation of a low molecular weight TLR7 ligand and α-mannose immobilized GNPs and its <i>in vitro</i> and <i>in vivo</i> immunostimulatory activities.</p>

    <p> Toll-like receptors (TLRs) are a family of type Ⅰ membrane proteins, classified to pattern recognition receptors (PRR). TLRs are expressed in cell surface or endosomal compartment of immune cells. TLRs are activated by recognition of pathogen specific components called pathogen-associated molecular patterns (PAMPs). Signal of TLRs induces the innate immune response and adaptive immune response. Therefore, TLRs play important roles in host defense system against viral or bacterial infection. In human, 10 kinds of TLR (TLR1 to TLR10) have been found. Each TLR recognizes different PAMP ; lipoprotein for TLR1/6 and TLR2/6 ; nucleic acid components for TLR3, -7, -8, and -9 ; lipopolysaccharide (LPS) for TLR4. TLRs are also activated by the specific endogenous danger signal (called damage-associated molecular pattern, DAMPs) produced by damaged tissue, which is thought to cause various chronic inflammatory diseases. On the other hand, low molecular weight TLR ligands have been found from natural and synthetic compounds, those can be candidates for development of chemical probes and drugs. In this review, focusing on TLR4, we describe recent progress in exploring TLR4 ligands with agonistic or antagonistic activity and our recent study on the structure-activity relationship of pyrimido [5,4-b] indole derivatives.</p>
