    <p>Reactive sulfur species (RSS) are present in root nodules; however, their role in symbiosis and the mechanisms underlying their production remain unclear. We herein investigated whether RSS produced by the cystathionine γ-lyase (CSE) of microsymbionts are involved in root nodule symbiosis. A <i>cse</i> mutant of <i>Mesorhizobium loti</i> exhibited the decreased production of hydrogen sulfide and other RSS. Although the <i>CSE</i> mutation of <i>M. loti</i> did not affect the early stages of symbiosis, <i>i.e.</i>, infection and nodulation, with <i>Lotus japonicus</i>, it reduced the nitrogenase activity of nodules and induced their early senescence. Additionally, changes in the production of sulfur compounds and an increase in reactive oxygen species (ROS) were observed in the infected cells of nodules induced by the <i>cse</i> mutants. The effects of CSE inhibitors in the <i>L. japonicus</i> rhizosphere on symbiosis with <i>M. loti</i> were also investigated. All three CSE inhibitors suppressed infection and nodulation by <i>M. loti</i> concomitant with decreased RSS levels and increased ROS and nitric oxide levels. Therefore, RSS derived from the CSE activity of both the microsymbiont and host plant are required for symbiosis, but function at different stages of symbiosis, possibly with crosstalk with other reactive mole­cular species.</p>

    <p>Root nodule symbiosis between legumes and rhizobia involves nitric oxide (NO) regulation by both the host plant and symbiotic rhizobia. However, the mechanisms by which the rhizobial control of NO affects root nodule symbiosis in <i>Lotus japonicus</i> are unknown. Therefore, we herein investigated the effects of enhanced NO removal by <i>Mesorhizobium loti</i> on symbiosis with <i>L. japonicus</i>. The <i>hmp</i> gene, which in <i>Sinorhizobium meliloti</i> encodes a flavohemoglobin involved in NO detoxification, was introduced into <i>M. loti</i> to generate a transconjugant with enhanced NO removal. The symbiotic phenotype of the transconjugant with <i>L. japonicus</i> was examined. The transconjugant showed delayed infection and higher nitrogenase activity in mature nodules than the wild type, whereas nodule senescence was normal. This result is in contrast to previous findings showing that enhanced NO removal in <i>L. japonicus</i> by class 1 phytoglobin affected nodule senescence. To evaluate differences in NO detoxification between <i>M. loti</i> and <i>L. japonicus</i>, NO localization in nodules was investigated. The enhanced expression of <i>class 1</i> <i>phytoglobin</i> in <i>L. japonicus</i> reduced the amount of NO not only in infected cells, but also in vascular bundles, whereas that of <i>hmp</i> in <i>M. loti</i> reduced the amount of NO in infected cells only. This difference suggests that NO detoxification by <i>M. loti</i> exerts different effects in symbiosis than that by <i>L. japonicus</i>.</p>

    Under water deficit conditions, the essential macronutrient nitrogen becomes limited as a result of reduced dissolved nitrogen and root nitrogen uptake. An elevated nitrogen level might be able to mitigate these effects, integrated with the idea of using nitric oxide as abiotic stress tolerant inducers. In this study, we evaluated the potential of using elevated nitrogen priming prior to water shortage to mitigate plant stress through nitric oxide accumulation. We grew rice plants in 300 mg L−1 nitrogen for 10 weeks, then we primed plants with four different nitrogen concentrations: 100, 300 (control), 500 and 1000 mg L−1 nitrogen prior to inducing water deficit conditions. Plants primed with 500 mg L−1 nitrogen possessed a higher photosynthetic rate, relative water content, electrolyte leakage and lipid peroxidation under water deficit conditions, compared to control plants. The induction of water deficit tolerance was supported with the activation of antioxidant defense system, induced by the accumulation of nitric oxide in leaves and roots of rice plants. We originally demonstrated the accumulation of nitric oxide in leaves of rice plants. The elevated nitrogen priming can be used to enhance water deficit tolerance in irrigated paddy fields, instead of nitric oxide donors.

    <p>Many abiotic stresses induce the generation of nitric oxide (NO) in plant tissues, where it functions as a signal molecule in stress responses. Plants modulate NO by oxidizing it to NO<sub>3</sub><sup>−</sup> with plant hemoglobin (GLB), because excess NO is toxic to cells. At least eight genes encoding GLB have been identified in soybean, in three clades: <i>GLB1</i>, <i>GLB2</i>, and <i>GLB3</i>. However, it is still unclear which <i>GLB</i> genes are responsible for NO regulation under abiotic stress in soybean. We exposed soybean roots to flooding, salt, and two NO donors—sodium pentacyanonitrosylferrate (III) dihydrate (SNP) and <i>S</i>-nitroso-<i>N</i>-acetyl-D,L-penicillamine (SNAP)—and analyzed expression of <i>GLB</i> genes. <i>GmGLB1</i>, one of two <i>GLB1</i> genes of soybean, significantly responded to both SNP and SNAP, and its induction was almost completely repressed by a NO scavenger, 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide. <i>GmGLB1</i> responded to flooding but not to salt, suggesting that it is responsible for NO regulation under NO-inducing abiotic stresses such as flooding. <i>GmGLB3</i>, one of two <i>GLB3</i> genes of soybean, did not respond to NO donors at all but did respond to flooding, at a lower level than <i>GmGLB1.</i> These results suggest that flooding induces not only NO but also unknown factor(s) that induce <i>GmGLB3</i> gene in soybean.</p>

    <p><i>Bradyrhizobium</i> sp. strain SUTN9-2 is a symbiotic and endophytic diazotrophic bacterium found in legume and rice plants and has the potential to promote growth. The present results revealed that SUTN9-2 underwent cell enlargement, increased its DNA content, and efficiently performed nitrogen fixation in response to rice extract. Some factors in rice extract induced the expression of cell cycle and nitrogen fixation genes. According to differentially expressed genes (DEGs) from the transcriptomic analysis, SUTN9-2 was affected by rice extract and the deletion of the <i>bclA</i> gene. The up-regulated DEGs encoding a class of oxidoreductases, which act with oxygen atoms and may have a role in controlling oxygen at an appropriate level for nitrogenase activity, followed by GroESL chaperonins are required for the function of nitrogenase. These results indicate that following its exposure to rice extract, nitrogen fixation by SUTN9-2 is induced by the collective effects of GroESL and oxidoreductases. The expression of the sensitivity to antimicrobial peptides transporter (<i>sapDF</i>) was also up-regulated, resulting in cell differentiation, even when <i>bclA</i> (<i>sapDF</i>) was mutated. This result implies similarities in the production of defensin-like antimicrobial peptides (DEFs) by rice and nodule-specific cysteine-rich (NCR) peptides in legume plants, which affect bacterial cell differentiation.</p>

    <p>The rhizobial type III secretion system secretes effector proteins into host plant cells, which may either promote or inhibit symbiosis with legumes. We herein demonstrated that the type III secretion system of <i>Bradyrhizobium</i> sp. SUTN9-2 obstructed symbiosis with <i>Lotus japonicus</i> Miyakojima, <i>L. japonicus</i> Gifu, and <i>Lotus burttii</i>. A mutant of SUTN9-2 that is unable to secrete effector proteins showed better nodulation and plant growth promotion than wild-type SUTN9-2 when paired with these <i>Lotus</i> spp. We propose that SUTN9-2 is a useful strain for understanding the mechanisms by which effector proteins obstruct symbiosis between <i>Bradyrhizobium</i> and <i>Lotus</i> spp.</p>

    <p>The <i>nifV</i> gene encodes homocitrate synthase, the enzyme that catalyzes the formation of homocitrate, which is essential for arranging the FeMo-cofactor in the catalytic center of nitrogenase. Some host plants, such as <i>Lotus japonicus</i>, supply homocitrate to their symbionts, in this case, <i>Mesorhizobium loti</i>, which lacks <i>nifV</i>. In contrast, <i>Bradyrhizobium</i> ORS285, a symbiont of <i>Aeschynomene</i> cross-inoculation (CI) groups 2 and 3, requires <i>nifV</i> for symbiosis with <i>Aeschynomene</i> species that belong to CI group 3, and some species belonging to CI group 2. However, it currently remains unclear whether rhizobial <i>nifV</i> is required for symbiosis with <i>Aeschynomene</i> species belonging to CI group 1 or with other legumes. We generated <i>nifV</i>-disruption (Δ<i>nifV</i>) mutants of two wide-host-range rhizobia, <i>Bradyrhizobium</i> SUTN9-2 and DOA9, to investigate whether they require <i>nifV</i> for symbiosis. Both Δ<i>nifV</i> mutant strains showed significantly less nitrogenase activity in a free-living state than the respective wild-type strains. The symbiotic phenotypes of SUTN9-2, DOA9, and their Δ<i>nifV</i> mutants were examined with four legumes, <i>Aeschynomene americana</i>, <i>Stylosanthes hamata</i>, <i>Indigofera tinctoria</i>, and <i>Desmodium tortuosum</i>. <i>nifV</i> was required for the efficient symbiosis of SUTN9-2 with <i>A. americana</i> (CI group 1), but not for that of DOA9. SUTN9-2 established symbiosis with all three other legumes; <i>nifV</i> was required for symbiosis with <i>I. tinctoria</i> and <i>D. tortuosum</i>. These results suggest that, in addition to <i>Aeschynomene</i> CI groups 2 and 3, CI group 1 and several other legumes require the rhizobial <i>nifV</i> for symbiosis.</p>

    <p>Aphids have a mutualistic relationship with the bacterial endosymbiont <i>Buchnera aphidicola</i>. We previously reported seven cysteine-rich peptides in the pea aphid <i>Acyrthosiphon pisum</i> and named them Bacteriocyte-specific Cysteine-Rich (BCR) peptides; these peptides are exclusively expressed in bacteriocytes, special aphid cells that harbor symbionts. Similar symbiotic organ-specific cysteine-rich peptides identified in the root nodules of leguminous plants are named Nodule-specific Cysteine-Rich (NCR) peptides. NCR peptides target rhizobia in the nodules and are essential for symbiotic nitrogen fixation. A BacA (membrane protein) mutant of <i>Sinorhizobium</i> is sensitive to NCR peptides and is unable to establish symbiosis. Based on the structural and expressional similarities between BCR peptides and NCR peptides, we hypothesized that aphid BCR peptides exhibit antimicrobial activity, similar to some NCR peptides. We herein synthesized BCR peptides and investigated their antimicrobial activities and effects on the bacterial membrane of <i>Escherichia coli</i>. The peptides BCR1, BCR3, BCR5, and BCR8 exhibited antimicrobial activities with increased membrane permeability. An <i>sbmA</i> mutant of <i>E. coli</i>, a homolog of <i>bacA</i> of <i>S. meliloti</i>, was more sensitive to BCR peptides than the wild type. Our results suggest that BCR peptides have properties that may be required to control the endosymbiont, similar to NCR peptides in legumes.</p>

    Leghemoglobins transport and deliver O2 to the symbiosomes inside legume nodules and are essential for nitrogen fixation. However, the roles of other hemoglobins (Hbs) in the rhizobia–legume symbiosis are unclear. Several Lotus japonicus mutants affecting LjGlb1-1, a nonsymbiotic class 1 Hb, have been used to study the function of this protein in symbiosis. Two TILLING alleles with single amino acid substitutions (A102V and E127K) and a LORE1 null allele with a retrotransposon insertion in the 5'-UTR (96642) were selected for phenotyping nodulation. Plants of all three mutant lines showed a decrease in long infection threads and nodules and an increase in incipient infection threads. About four hours after inoculation, the roots of mutant plants exhibited a greater transient accumulation of nitric oxide (NO) than did the wild-type roots; nevertheless, in vitro NO dioxygenase activities of the wild-type, A102V, and E127K proteins were similar, suggesting that the mutated proteins are not fully functional in vivo. The expression of LjGlb1-1, but not of the other class 1 Hb of L. japonicus (LjGlb1-2), was affected during infection of wild-type roots, further supporting a specific role of LjGlb1-1. In conclusion, the LjGlb1-1 mutants reveal that this protein is required during rhizobial infection and regulates NO levels.

    Phytohormone abscisic acid (ABA) inhibits root nodule formation of leguminous plants. LjGlu1, a β-1,3-glucanase gene of Lotus japonicus, has been identified as an ABA responsive gene. RNA interference of LjGlu1 increased nodule number. This suggests that LjGlu1 is involved in the regulation of nodule formation. Host legumes control nodule number by autoregulation of nodulation (AON), in which the presence of existing root nodules inhibits further nodulation. For further characterization of LjGlu1, we focused on the expression of LjGlu1 in relation to AON. In a split-root system, LjGlu1 expression peaked when AON was fully induced. Hairy roots transformed with LjCLE-RS1, a gene that induces AON, were generated. Expression of LjGlu1 was greater in the transgenic roots than in untransformed roots. LjGlu1 was not induced in a hypernodulating mutant inoculated with Mesorhizobium loti. These results suggest that the expression of LjGlu1 is involved in the system of AON. However, neither hypernodulation nor enlarged nodulation zone was observed on the transgenic hairy roots carrying LjGlu1-RNAi, suggesting that LjGlu1 is not a key player of AON. Recombinant LjGlu1 showed endo-β-1,3-glucanase activity. LjGlu1-mOrange fusion protein suggested that LjGlu1 associated with M. loti on the root hairs. Exogenous β-1,3-glucanase inhibited infection thread formation by both the wild type and the mutant, and nodule numbers were reduced. These results suggest that LjGlu1 is expressed in response to M. loti infection and functions outside root tissues, resulting in the inhibition of infection.

    Hemoglobins (Hbs) are ubiquitous in plants and can be grouped into classes 1, 2, and class 3 based on phylogenetic analyses and biochemical properties. Class 1 Hbs are expressed in most plant organs, including seeds, roots, leaves, and nodules, where they may perform multiple functions by modulating nitric oxide (NO) levels. Because NO inhibits nitrogenase activity and is generated inside the nodules, the NO scavenging activity of class 1 Hbs may protect symbiotic nitrogen fixation. However, it is unclear whether class 1 Hbs are also important during the infection process. To investigate this, we produced and selected several mutant lines of the LjGlb1-1 gene of L. japonicus. These were a TILLING line with a single-site mutation (A102V) and a null mutant LORE1 line (96642) having a retrotransposon insertion in the 5'-UTR. The mutants were smaller, produced fewer nodules, and had lower nitrogenase activity. The use of a DsRed-tagged rhizobium strain revealed inhibition of infection thread (IT) elongation in the mutant roots, with an increase in the number of new ITs and a decrease in the number of long ITs per root length. Four hours after inoculation with M. loti, the roots of the mutant lines exhibited a greater transient accumulation of NO than did the WT roots. Nevertheless, the spectra and NO dioxygenase activities of the recombinant WT and A102V proteins were similar, suggesting that the mutated protein is unstable and/or not fully functional in vivo. The expression of LjGlb1-1, but not of the other class 1 Hb gene of L. japonicus (LjGlb1-2), was affected during infection of WT roots, further supporting a specific role of LjGlb1-1. These results show that LjGlb1-1 is required during rhizobial infection and regulates NO levels.

    Hemoglobin (Hb) is ubiquitous in the plant kingdom. Plant Hbs are classified into class 1, class 2 and class 3, based on their amino acid sequences. The function of the class 3 Hb is obscure. Class 2 Hb of leguminous plants, i.e. leghemoglobin (Lb), controls oxygen partial pressure in the root nodule and is essential for symbiotic nitrogen fixation. Class 1 Hb is present in the whole plant and is involved in the regulation of various physiological functions of the plant by interacting with nitric oxide (NO). Because NO is known to be an inhibiter of nitrogenase activity and is detectable inside the nodules, function of NO scavenging activity of class 1 Hb may be involved in the symbiotic nitrogen fixation. However, it is not clear whether the NO scavenging activity of class 1 Hb has any effect on the symbiotic nitrogen fixation.
    We established three mutant lines, two lines of amino acid substitution and a low expression line caused by LORE1 insertion, of class1 Hb gene (LjGlb1-1) of Lotus japonicus Gifu. NO levels in the roots of these mutant lines were higher than that of native Gifu. The mutant lines showed inhibition of infection thread elongation, decreased number of nodules and lowered nitrogenase activity. These results show that LjGlb1-1 regulates NO levels and is required during rhizobial infection. NO is produced in the nodules by senescence and by flooding. NO regulatory function may contribute to repress the stresses caused by senescence and by flooding. We established overexpression lines of class 1 Hb gene (LjGlb1-1) of L. japonicus Gifu. The LjGlb1-1 overexpression lines showed enhanced nitrogenase activity and reduced amount of NO in the roots. Nodule senescence and flood-resistant were assessed by the color of nodules, nitrogenase activity, microscopic observation and NO content of the nodules. The LjGlb1-1 overexpression lines showed delay on nodule senescence and higher flood-resistant compared with those of native Gifu. These results suggest that the control of NO by LjGlb1-1 are also involved in the life and flood resistance of the nodules. The expression of two genes, which showed higher expression in aged nodules, was not enhanced by flooding. Aging and flooding induce NO production but may affect on the function of nodules in the different way.
