    Massa Medicata Fermentata(MMF)はAspergillus属やRhizopus属による微生物発酵でコムギと数種のハーブから調製されるので、微生物発酵の影響について検討した。National Standard for Chinese Patent Drugsに従ってMMFの原材料を混合し、Aspergillus oryzae培養で調製(MMF-A)またはRhizopus oryzae培養で調製(MMF-R)した。MMFの調製により酵素活性は上昇した。MMF-AはMMF-Rよりもα-アミラーゼとリパーゼの活性が高かった。プロテアーゼ活性はMMF-Rがわずかに高かった。テルペノイドは発酵で低下したが、2,3-ブタンジオール、アセトイン、グアヤコールはMMFだけで検出された。1-オクテン-3-オールのC8化合物はMMF-RよりもMMF-Aで高く、4-ビニルグアヤコールとピラジンの芳香族化合物はMMF-Rで高かった。

    Aim: Massa Medicata Fementata (MMF) is a crude drug used in East Asia to treat anorexia and dyspepsia. It is prepared from wheat and several herbs through microbial fermentation using Aspergillus sp. and Rhizopus sp. There is great difference in the quality of commercial MMF, and the microbes of MMF are suggested to affect its quality. We investigated the effects of microbial fermentation on the quality of MMF. Methods: Raw materials of MMF were mixed according to the ratio listed in the National Standard for Chinese Patent Drugs, and MMF was prepared using pure cultures of Aspergillus oryzae or Rhizopus oryzae. Digestive enzyme activities (α-amylase, protease, and lipase) and volatile compounds were measured using an analytical kit and GC–MS, respectively. Results: Enzyme activity increased in MMF. MMF prepared with A. oryzae (MMF-A) showed higher α-amylase and lipase activities than that prepared with R. oryzae (MMF-R). Protease activity was marginally higher in MMF-R than in MMF-A. GC–MS analysis revealed that terpenoids decreased with fermentation; however, 2,3-butanediol, acetoin, and guaiacol were detected in MMF only. C8 compounds such as 1-octen-3-ol were higher in MMF-A than MMF-R; however, aromatic compounds such as 4-vinylguaiacol and pyrazines were higher in MMF-R than MMF-A. Conclusion: Microbial fermentation contributes to increased enzyme activity and changes in MMF volatiles. These properties of MMF were considerably affected by the microbes used, and it is proposed in this study that it is important to have microbial control in the production of commercial MMF.

    <p>This study investigated genetic polymorphism related to anthocyanin composition variation in lisianthus (<i>Eustoma grandiflorum</i>) flower petals. Three different bands were detected by genomic polymerase chain reaction using specific primers based on the reported flavonoid 3',5'-hydroxylase (<i>F3'5'H</i>) gene sequence. Our genetic study revealed that they were allelic. They were named <i>F3'5'H1-1</i>, <i>F3'5'H1-2</i>, and <i>f3'5'h1-2</i>. <i>F3'5'H1-1</i> and <i>F3'5'H1-2</i> differed in the length of the intron. Allele <i>f3'5'h1-2</i> possessed a retrotransposon insertion in the first exon, but otherwise, its gene sequence was almost identical to that of <i>F3'5'H1-2</i>. This retrotransposon is a Ty1-<i>copia</i> type retrotransposon and was designated as the retrotransposon of <i>Eustoma grandiflorum</i> 1 (<i>rTeg1</i>). The mauve flower line ‘ME’ and red-purple flower line ‘PRP’ were homozygous for <i>F3'5'H1-1</i> and mostly accumulated cyanidin, with a small amount of delphinidin, in flower petals (the Cy major phenotype). The violet flower line ‘RV’ was homozygous for <i>F3'5'H1-2</i> and accumulated only delphinidin (the Dp major phenotype). The pink-red flower line ‘AK’ was homozygous for<i> f3'5'h1-2</i>, accumulated pelargonidin, and lacked delphinidin (the Pg major-non Dp phenotype). Segregation analysis of the F<sub>2</sub> population from ‘ME’ × ‘RV’ revealed that <i>F3'5'H1-2</i> was associated with the Dp major phenotype, and the delphinidin major trait was dominant in the Cy major phenotype. All the Cy major phenotype plants in the F<sub>2</sub> population possessed homozygous <i>F3'5'H1-1</i>. Genetic analysis of the F<sub>2</sub> population from the cross between delphinidin-containing and delphinidin-lacking strains revealed that the homozygous <i>f3'5'h1-2</i> genotype co-segregated with the Pg major-non Dp phenotype, which was recessive to the delphinidin-containing phenotypes. Expression analysis of the <i>F3'5'H</i> gene demonstrated that an abnormal mRNA was transcribed from the <i>F3'5'H</i> gene in the homozygous <i>f3'5'h1-2</i> line by the insertion of <i>rTeg1</i>. Therefore, these <i>F3'5'H</i> gene polymorphisms can be used as DNA markers for breeding lisianthus based on flower color.</p>

    <p>Novel mutant alleles related to isoflavone content are useful for breeding programs to improve the disease resistance and nutritional content of soybean. However, identification of mutant alleles from high-density mutant libraries is expensive and time-consuming because soybean has a large, complicated genome. Here, we identified the gene responsible for increased genistein-to-daidzein ratio in seed of the mutant line F333ES017D9. For this purpose, we used a time- and cost-effective approach based on selective genotyping of a small number of F<sub>2</sub> plants showing the mutant phenotype with nearest-neighboring-nucleotide substitution–high-resolution melting analysis markers, followed by alignment of short reads obtained by next-generation sequencing analysis with the identified locus. In the mutant line, <i>GmCHR5</i> harbored a single-base deletion that caused a change in the substrate flow in the isoflavone biosynthetic pathway towards genistein. Mutated <i>GmCHR5</i> was expressed at a lower level during seed development than wild-type <i>GmCHR5</i>. Ectopic overexpression of <i>GmCHR5</i> increased the production of daidzein derivatives in both the wild-type and mutant plants. The present strategy will be useful for accelerating identification of mutant alleles responsible for traits of interest in agronomically important crops.</p>

    <p>The optimum temperature during fruit ripening period of passion fruit (<i>Passiflora edulis</i>) was elucidated in the present study. 'Summer Queen' (<i>P. edulis</i> × <i>P. edulis</i> f. <i>flavicarpa</i>) and 'Ruby Star' (<i>P. edulis</i> × <i>P. edulis</i> f. <i>flavicarpa</i>) showed a greenish fruit peel and low total soluble solids/titratable acidity (TSS/TA) ratio at 15°C. In 'Summer Queen', a high °Brix, low titratable acidity, and high TSS/TA ratio were observed at 25°C. In 'Ruby Star', the peel coloration was highly favorable at 25°C. In both cultivars, the peel tended to be yellowish and °Brix of the juice was low at 35°C. These results indicate that the optimal temperature for passion fruit ripening was 25°C. In addition, we elucidated the varietal differences in the ripening characteristics of fruit on and off the vine.</p>

    <p>Soybean isoflavones are functionally important secondary metabolites that are mainly accumulated in seeds. Their biosynthetic processes are regulated coordinately at the transcriptional level; however, screening systems for key transcription factors (TFs) are limited. Here we developed a combination screening system comprising a simple agroinfiltration assay and a robust hairy root transformation assay. First, we screened for candidate MYB TFs that could activate the promoters of the chalcone synthase (CHS) gene <i>GmCHS8</i> and the isoflavone synthase (IFS) genes <i>GmIFS1</i> and <i>GmIFS2</i> in the isoflavone biosynthetic pathway. In the agroinfiltration assay, we co-transformed a <i>LjUbi</i> (<i>Lotus japonicus</i> <i>polyubiquitin</i> gene) promoter-fused <i>MYB</i> gene with target promoter-fused <i>GUS</i> (β-glucuronidase) gene constructs, and identified three genes (<i>GmMYB102</i>, <i>GmMYB280</i>, and <i>GmMYB502</i>) as candidate regulators of isoflavone biosynthesis. We then evaluated the functional regulatory role of identified three MYB genes in isoflavone biosynthesis using hairy roots transformation assay in soybean for the accumulation of isoflavones. Three candidate MYB genes showed an increased accumulation of total isoflavones in hairy root transgenic lines. Accumulation of total isoflavones in the three MYB-overexpressing lines was approximately 2-to 4-folds more than that in the vector control, confirming their possible role to regulate isoflavone biosynthesis. However, the significant accumulation of authentic <i>GmCHS8</i>, <i>GmIFS1</i>, and <i>GmIFS2</i> transcripts could not be observed except for the GmMYB502-overexpressing line. Therefore, the analysis of isoflavone accumulation in transgenic hairy root was effective for evaluation of transactivation activity of MYB TFs for isoflavone biosynthetic genes. Our results demonstrate a simple and robust system that can potentially identify the function of orphan TFs in diverse plant metabolic pathways.</p>

    <p>Shimatogarashi (<i>Capsicum frutescens</i>) is a typical chili pepper domesticated in southern Japan. Important traits of Shimatogarashi peppers, such as color; proportion of organic acids, capsaicinoids, and aromatic compounds; and antioxidant activity in three stages of maturity (green (immature), orange (turning), and red (mature) stages) were characterized. The results indicated that the concentration of organic acids, including ascorbic, citric, and malic acid, increased during ripening. In addition, the amount of capsaicinoids, which are responsible for the pungent taste of chili peppers, increased as the fruit matured to the orange and red stages. The volatile compound profile of Shimatogarashi was dominated by the presence of esters, which mainly contributed to fruity notes. The total amount of volatile compounds analyzed by gas chromatography-headspace solid-phase microextraction (GC-HS-SPME), especially esters, decreased as the fruit changed in color from green to red. This was in contrast to the amount of terpenoids, especially limonene, which increased at the red stage, denoting a change in flavor from fruity to a more citrus-like aroma. Based on the total phenolic content (TPC), the oxygen radical absorbance capacity (ORAC) and the diphenylpicrylhydrazyl (DPPH) free radical method, the antioxidant capacity of Shimatogarashi showed an increase at the mature red stage. However, while the red stage showed higher pungency and antioxidant capacity as well as an attractive color, the results of aromatic compound analysis revealed that the immature green stage had the advantages of having pleasant fruity smell, making it suitable for use in condiments.</p>
