    A mass production test of <i>tsukuritake </i>mushrooms (<i>Agaricus bisporus</i>) that can be cultivated in compost was conducted to develop a new utilization method for sewage sludge compost. The fruit body yield was 1350 &plusmn; 230 g / container in the test plots using sewage sludge compost/cow manure, which was comparable to the control plot using horse manure compost. In the test plots using sewage sludge compost, the amount of free amino acids in fruit bodies increased by 50-70 % compared to the control plots. Furthermore, heavy metal analysis of fruiting bodies showed the detection of Cd and T-Hg in fruit bodies in the test plots using sewage sludge compost. Judging from the acceptable values for agricultural products and the content of heavy metals in domestically grown mushrooms, however, the values of the fruit bodies cultivated in this test were judged to have no effect on health. The waste fungus bed was subjected to a cultivation test of komatsuna (<i>Brassica rapa </i>var. <i>perviridis</i>), and it was found that the mushroom waste bed containing sewage sludge compost and cow dung compost especially grew more than the section with chemical fertilizer application and could be used as a partial substitute for fertilizer.

    <p>The uranium (U) concentrations of the agricultural and non-agricultural soils in four areas (Isa, Nagashima, Nansatsu, and Osumi) in Kagoshima Prefecture were determined. The U concentrations of the agricultural soils were higher than those of the non-agricultural soils in all areas. The U concentrations were positively correlated with the P concentrations in the soils, which suggested a link between long-term phosphate fertilizer application and U accumulation in the soils. The amounts of U enriched in the agricultural soil surfaces (0-15 cm depth) in the Isa, Nagashima, Nansatsu, and Osumi areas, as calculated from the comparison with the non-agricultural soils, were estimated to be 47-318, 25-343, 61-226, and 47-185 mg/m<sup>2</sup>, respectively. This demonstrated that the accumulation of U occurred in the agricultural soils in these areas. The commercially available superphosphate, magnesium multi-phosphate, fused magnesium phosphate, animal waste composts, and sewage sludge compost in these areas contained 51.6, 22.2, 10.6, 0.88-1.07, and 1.56 μg/g of U, respectively, which showed that these fertilizer materials could become sources of U in agricultural soils. According to the calculation with reference to the amount of fertilizer applied, it was estimated that the amounts of U added from fertilizers were 2.7-3.0 mg/m<sup>2</sup> in rice paddy fields and 8.6-9.2 mg/m<sup>2</sup> in Chinese cabbage fields per cropping system. These results indicated that the amounts of U input derived from fertilizers per cropping system differed greatly depending on the cultivated crop.</p>

    <p>PCR clamping by locked nucleic acid (LNA) oligonucleotides is an effective technique for selectively amplifying the community SSU rRNA genes of plant–associated bacteria. However, the original primer set often shows low amplification efficiency. In order to improve this efficiency, new primers were designed at positions to compete with LNA oligonucleotides. Three new sets displayed higher amplification efficiencies than the original; however, efficiency varied among the primer sets. Two new sets appeared to be available in consideration of bacterial profiles by next-generation sequencing. One new set, KU63f and KU1494r, may be applicable to the selective gene amplification of plant-associated bacteria.</p>

    &nbsp;In this study, we carried out the button mushroom cultivation tests on media covered with peat moss which induces primordia in order to improve the mushroom yield. By the peat moss's properties of high density and low water absorption rate, the yield increased 1.5-1.7 times compared to the past test results, which achieved the target value (150-250 kg/1000kg compost) for management of button mushroom production. Since the button mushroom requires a relatively long cultivation period, it greatly influences on the medium composition. It is presumed that composition of fruit bodies will change depending on the difference of medium composition. Therefore, the effect of the differences in harvest time on the composition of fruit bodies was investigated. The composition of fruit bodies varied due to the occurring time. We found that the amount of protein increased in the fruit bodies occurred later. The mushroom waste beds were used for the cultivation of leaf vegetables by the general application method of compost. The growth of Komatsuna (Japanese mustard spinach) somewhat diminished, but the growth of Chingensai (Bok choy) were equal or higher compared to the control. These results led us to conclude that the waste bed were usable for agricultural purpose.

    <p>The effect of methane fermentation liquid obtained from livestock waste slurry (MFL) on the activity of root-knot nematodes (<i>Meloidogyne</i> spp.) was evaluated. On the day after preparation, MFL contained 1.86 g L<sup>–1</sup> of ammonium nitrogen which is reported to have nematocidal activity. However, the ammonium nitrogen content decreased with the lapse of storage time. More than 90% of the population of <i>M.incognia</i> was immobilized by adding the five-times-diluted MLF solution to a suspension of <i>M.incognita</i>. This immobilization effect was also observed in other <i>Meloidogyne</i> spp.: <i>M. arenaria</i> Honshu-type, <i>M. arenaria</i> Okinawa-type, <i>M.hapla</i>, <i>M.javanica</i> and <i>M.microcephalus</i>. The population of <i>M.incognita</i> in soil was significantly reduced by addition of 120 mL kg<sup>–1</sup> (equivalent to 60% of maximum water-holding capacity) MLF to soil in Sand-dune Regosol infested with <i>M.incognita</i>. However, this effect was not observed clearly in Gray Lowland soil and Andosol. The amount of egg masses produced by <i>M.incognita</i> was reduced to less than 25% of that in the control by applying 70 mL kg<sup>–1</sup> MLF (containing 200 mg NH<sub>4</sub><sup>+</sup>-N) to the soil of nematode-infected Sand-dune Regosol in the pot experiment with tomato seedling.</p>
